The principle of cell fragmentation by a Constant cell disruptor 
The Constant Systems high-pressure cell disruptor is specifically developed for biotechnology users, taking into account the characteristics of biological samples in design. It has unique designs for sample cooling, CIP, and SIP. The principle of the Constant Systems high-pressure disruptor is to use high-pressure extrusion of samples to pass through a fixed small nozzle at high speed, and then collide with the impact target to break the cells.

The high-pressure piston moves downwards, and due to gravity, the sample is sucked into the high-pressure sleeve

 

Once the sample enters the high-pressure chamber, the piston accelerates at high speed to close the inlet valve and allows the sample to pass through the fixed nozzle at high speed. The sample pressure can reach 40kpsi (2700 bar), and the sample quickly transitions from the high-pressure zone to the low-pressure zone, causing cell fragmentation

 

The sample hits the impact target and flows through the cooling surface. The cooled sample flows out of the outlet due to gravity and enters the lower cycle


Temperature control                                                                        

By using a cooling jacket to cool the sample during the disrupting process, the temperature of the disrupted sample can be well controlled, thereby avoiding the deactivation of active substances.

Technical characteristics of Constant cell fragmentation equipment

Constant Systems can generate a pressure of up to 40Kpsi (2700 bar), with good disrupting effect. One disrupting can achieve very high disrupting efficiency, avoiding multiple disrupting that may cause fine fragments and denaturation and deactivation of active substances.

Constant Systems disrupting head has a cooling jacket design, which can effectively control the temperature of disrupted samples; Enable the sample to be cooled in a timely manner during the disrupting process, thereby avoiding the deactivation of active substances due to temperature rise in the disrupted sample; In addition, the sample storage tank can also be equipped with a jacket for sample pre cooling.

Constant Systems adopts a fixed nozzle design, which has good stability; Overcoming the disadvantages of valve design that relies on regulating valves to regulate disrupting pressure, such as poor pressure stability, poor reproducibility, and easy valve failure during the disrupting process.

Constant Systems uses hydraulic systems to generate high pressure, with stable pressure and good reproducibility.

The Constant Systems high-pressure cell disruptor has a dead volume of less than 1ml (at least less than 0.1ml) and no minimum sample processing capacity limit.

Constant Systems can process samples with a concentration of up to 30% bacterial concentration, resulting in high concentration of broken sample products, making it convenient for downstream separation and purification operations; Can handle filamentous bacterial cells.

Constant Systems TS, B, and C series optional peristaltic pumps for fully automatic injection and fully enclosed operation, to avoid affecting or being affected by the environment.

The operation and maintenance are simple, and CIP and SIP are very convenient. Specific CIP and SIP designs can be selected, which comply with GMP and FDA standards.
