Applitech Sheet-Like Microcarrier Cell Counting Solution
Sheet-like microcarrier cell culture offers significant advantages in increasing yield, simplifying operations, reducing overall production costs, and minimizing contamination. It is particularly suitable for large-scale biopharmaceutical and vaccine production.
Currently, the monitoring of cell status in sheet-like microcarrier cultures typically involves detaching and digesting cells from the carrier using trypsin (crystal violet) for counting. This method has the following drawbacks:
- Multiple steps and time-consuming process
- Centrifugation easily causes cell clumping
- Incomplete trypsin digestion leads to partial cell release
- Microcarrier interference affects cell counting results (bright field)
Applitech provides the Countleader FL 1000 or FL 100 cell counter, which lyses cell membranes using a lysis reagent (Reagent A+B) to release intracellular nuclei. With fluorescent staining of nuclear DNA, it enables accurate cell counting and monitoring of cell growth status.

Case Study: MRC-5 Cell Culture on Sheet-Like Carriers three sheet-like carriers were taken for lysis and counting, and the average cell count per carrier was compared.

Results:
After lysing with 5 mL of Reagent A for 10 minutes, the obtained cell count was significantly higher—several times greater than that of trypsin digestion. This method provides a more accurate reflection of the actual cell culture conditions in the bioreactor.
